چکیده

Whitefly (Bemisia tabaci) is a polyphagous insect that causes huge damage in several horticultural crops, including tomato, by sucking nutrients from the phloem and transmitting viruses. Whiteflies are particularly difficult to manage use of chemicals remains common practice, which development insecticidal resistance. Thus, there considerable interest introduction resistance whitefly classical molecular breeding. Here, we explored concept using an RNA interference construct silence v-ATPase gene whiteflies interacting with transgenic tomato plants express siRNA molecules corresponding fragment B. tabaci vATPase. PCR analyses revealed presence both ΔATPase nptII transgenes all lines. expressing lines were challenged against mortality rate 57.1% line 4.4.1, while control was 7.6%. Mortality 2nd instar nymphs higher on 3rd slightly longer than plants. Although attraction insects not significantly different between treatments, number eggs laid lower, compared controls. RT-qPCR decreased expression level endogenous feeding No unexpected effect observed non-target Myzus persicae or Tuta absoluta. Results presented here may form foundation for generation elite varieties resistant whitefly, devastating pest. Genetically modified generated vATPase small interfering RNA. Transgenic tolerant no

Tissue-specific gene silencing by RNA interference in the whitefly Bemisia tabaci (Gennadius).

The hemipteran whitefly Bemisia tabaci (Gennadius) species complex and the plant viruses they transmit pose major constraints to vegetable and fiber production, worldwide. The whitefly tissue- and developmental-specific gene expression has not been exhaustively studied despite its economic importance. In 2002, a functional genomic project was initiated, which generated several thousands express...

RNA Interference based Approach to Down Regulate Osmoregulators of Whitefly (Bemisia tabaci): Potential Technology for the Control of Whitefly

Over the past decade RNA interference (RNAi) technology has emerged as a successful tool not only for functional genomics, but in planta expression of short interfering RNAs (siRNAs) that could offer great potential for insect pest management. The diet of insects feeding exclusively on phloem sieves contains water and sugars as main components, and the uptake of the liquid food greatly depends ...

siRNA Machinery in Whitefly (Bemisia tabaci)

BACKGROUND RNA interference has been emerged as an utmost tool for the control of sap sucking insect pests. Systemic response is necessary to control them in field condition. Whitefly is observed to be more prone to siRNA in recent studies, however the siRNA machinery and mechanism is not well established. METHODOLOGY/PRINCIPAL FINDINGS To identify the core siRNA machinery, we curated transcr...

RNA interference-mediated knockdown of the hydroxyacid-oxoacid transhydrogenase gene decreases thiamethoxam resistance in adults of the whitefly Bemisia tabaci

Bemisia tabaci has developed a high level of resistance to thiamethoxam, a second generation neonicotinoid insecticide that has been widely used to control this pest. In this study, we investigated whether hydroxyacid-oxoacid transhydrogenase (HOT) is involved in resistance to the neonicotinoid insecticide thiamethoxam in the whitefly. We cloned the full-length gene that encodes HOT in B. tabac...

Replication of Tomato Yellow Leaf Curl Virus in Its Whitefly Vector, Bemisia tabaci.

UNLABELLED Tomato yellow leaf curl virus (TYLCV) is a begomovirus transmitted exclusively by the whitefly Bemisia tabaci in a persistent, circulative manner. Replication of TYLCV in its vector remains controversial, and thus far, the virus has been considered to be nonpropagative. Following 8 h of acquisition on TYLCV-infected tomato plants or purified virions and then transfer to non-TYLCV-hos...
